The growth and differentiation of normal cells are controlled by protein-tyrosine kinases, which serve as receptors for a wide variety external signals. Small protein modules called Src homology 2 (SH2) SH3 domains mediate protein-protein interactions in signaling pathways that triggered tyrosine kinases. SH2 domain, module around 100 amino acids, is present kinase targets within the cell. recruited to activated autophosphorylated factor directly recognizing phosphorylation sites. Growth other phosphoproteins have short phosphotyrosine (pTyr)-containing sequences bound domains. distinct element approximately 50 residues recognizes proline-rich hydrophobic-amino-acid-containing regions, frequently found SH2-containing proteins. Tyrosine kinases can be coupled downstream with SH3-binding sites proteins acting adaptors.
